Yoh is quite pretty, but constantly reveals new ways to suck and get beaten up. Mockingbird looks like he should be about 12, and his childishness and awkward gender confusion aren't going to fly in the mainstream, I think. Crow... ok, Crow is pretty awesome, but Owl is kind of just sad, and Genkaku has no eyebrows and is obnoxious.
Most of the others wind up kicking the bucket way too quickly to matter (Rokuro, anime Ueshima), or are introduced with the understanding/requirement that they eventually die (Shishito, Tamaki, Rinchiro). And a bunch are just odd looking (Genkaku, Tamaki, Rinchiro, tiger dude, Shishito, Mockingbird) or underdeveloped (manga Ueshima)
I think it'll be for guys what Deathnote is for girls (since most of the hotness is in the females, like Shiro, Makina, and Karako) but will fail to be for girls what Deathnote is for guys (sheer awesome appealing to their demographic). It'll attract the same girls who were on team Ryuk. And I admit I'm generalizing as everyone being straight and/or adhering to their stereotypes, since we're talking mainstream, and that's how mainstream needs to be talked. It could also be compared to Bleach, but it's still missing the handsome men, and the violence is marginally more realistic, and considerably more gory.
I personally love Deadman Wonderland, and would adore if it got big. As long as it doesn't get all the Hetalia fans with the bad reputation. Luckily, the two are pretty incompatible...

My guess would be Panty & Stocking and Durarara, and if I knew anything about Homestuck, I might have something to say on that matter, too. It doesn't matter if Durarara is too complicated; lots of people watch for the pretty pictures and/or big explosions (and no, I'm not being mean. It's true. That's what one of my dear friends watches for) and Deathnote was complicated, too, but still caught on massively.
It doesn't hurt that it's dubbed, either. I think that the big one will be something (probably) dubbed and shonen and full of awesome (Like Deathnote or Bleach) or ridiculous shojo (Like Ouran or Hetalia)*. Hence Panty & Stocking and Durarara.
Anything that has potential to make a good video game also has promise, and I think Drrr and Panty & Stocking both fulfill that. Heck, Panty & Stocking practically is a videogame, as it's described.
*Please don't assume I mean that shonen is awesome and shojo is dumb. I don't. I mean a ridiculous shojo is going to make it more than an awesome one (see Ouran and Hetalia versus ...something even I can't think of), and an awesome shonen will work better than a ridiculous one (Bleach, Deathnote versus ... something else I can't even think of. Ridiculous shonen is usually pretty ecchi). There are, of course, exceptions, like Vampire Knight (but that was romance - LOTS of romance - plus awesome explosions and stuff) but they are less common and generally slightly less big.
